FOXO4-DRI is a D-retro-inverso peptide modeled on the FOXO4 transcription factor’s p53 binding domain. de Keizer and colleagues at the Hubrecht Institute demonstrated that disrupting the FOXO4-p53 interaction triggers apoptosis selectively in senescent cells while sparing healthy cells.1
In aged and chemotherapy-aged mice, FOXO4-DRI improved fitness, fur density, and renal function — the most striking senolytic peptide data published to date.1 Human clinical data is sparse; we prescribe it as an experimental longevity peptide for select clients who understand its early-stage status.

Mechanism — Senescent cell selectivity
Disrupts FOXO4-p53 binding, releasing p53 to drive apoptosis specifically in senescent cells where FOXO4 is elevated.1

Senolytic mechanism. Selective elimination of senescent cells documented in aged mouse models.1
Renal function. Recovery of renal function markers in chemotherapy-aged mice.

Side effects: Limited human data. Theoretical injection-site reactions, fatigue from senescent-cell clearance, occasional flushing.
